疏风解毒胶囊对于急性支气管炎大鼠模型的保护作用

摘要:目的:研究疏风解毒胶囊对急性支气管炎大鼠模型的保护作用,并探讨其对于炎性因子的分泌影响。方法:体重g左右的SPF级雄性SD大鼠60只,随机均分为对照(A)组、模型(B)组、疏风解毒胶囊治疗(C)组、地塞米松治疗(D)组。除A组外,余3组每天烟雾刺激2次,早晚各1次,共1周,制备成急性支气管炎大鼠模型。疏风解毒胶囊治疗组每天灌胃给药疏风解毒胶囊mg/kg、地塞米松治疗组每天腹腔注射地塞米松5mg/kg,于成模后第1天、3天、5天每组各处死5只,取其左主支气管组织作以下测试:HE染色病理学观察;气管组织IL-1β、IL-6免疫荧光标记后激光共聚焦显影。经颈静脉采血行酶联免疫吸附法(Elisa)检测IL-1β、TNF-α、Cxcl10及Ccl3表达水平。结果:模型组大鼠呼吸道上皮炎性反应明显,其呼吸道上皮组织在成模后第1天时出现明显的渗出炎性反应,第3、5天时呼吸道上皮组织炎性反应达到高峰伴有明显的纤毛紊乱,部分组织坏死脱落。模型组大鼠血清的IL-1β、IL-6、TNF-α、Cxcl10及Ccl3水平较对照组有明显升高(p0.05)。疏风解毒胶囊治疗组可明显缓解大鼠急性呼吸道上皮组织的损伤,其呼吸道上皮组织病理与模型组比较,成模后第1、3、5天时呼吸道上皮组织的炎性渗出程度较轻。疏风解毒胶囊治疗组大鼠血清的IL-1β、IL-6、TNF-α、Cxcl10及Ccl3水平较模型组有明显下降(p0.05)。疏风解毒胶囊治疗组与地塞米松治疗组相比较,两组大鼠血清的IL-1β、IL-6、TNF-α、Cxcl10及Ccl3水平无明显差异(p0.05)。结论:疏风解毒胶囊可减轻急性支气管炎大鼠模型呼吸道上皮细胞炎性因子的分泌,缓解大鼠的急性呼吸道上皮炎性反应。

Abstract:ObjectiveToobservetheprotectiveeffectsofShufengJieduCapsule(SFJDC)onratmodelofacutebronchitisandtoexploreitsinfluenceintheofinflammatoryfactorsinrespiratoryepithelium.MethodsSixtySPFSDratswererandomlydividedinto4groups:control(A)group.control(B)group.control(C)group.control(D)group.With15ratsineachgroup.ExceptfortheAgrouptheothersweresubjectedtosmoketwiceadayforatotalofweek.inordertotheratmodelofacutebronchitis,TheCgroupwasgiventheadministrationSFJDCbyoralgavageatadoseofmg/kg,andtheDgroupwasgiventheadministrationofDexamethasonebyintraperitonealinjectionatadoseof5mg/kg.At1stday,3rddayand5thdayafterthemodeling.5ratsrandomlychosenfromeachgroupweresacrificedtoharvesttheirleftbronchustissues.HEstainingwasusedtoobservethepathologicalmanifestation.TheconfocallaserscanningmicroscopetechnologywasusedtoobservethefluorescencelabelingofIL-1β、IL-6.ElisawasusedtodetectthelevelsofIL-1β、IL-6、TNF-α、Cxcl10andCcl3injugularveinblood.Results:theresultsofpathologicalmanifestationshowedthattheinflammatoryinfiltrationwasobviouslyobservedintherespiratorytissueinthegroupBwhentheratmodelofacutebronchitiswassuccessfullyestablishedfor1day.Wheretheinflammatoryinfiltrationreachedapeak.
